Digitization is often described in bit depth. The bit depth describes the number of grey values in an image, or the number of different tones which can be distinguished. A high dynamic range in an image requires a higher bit depth in order to be able to see all the details in the grey values of the image.

When capturing microscopy images if digital gain is used too much it will result in dark areas in the image. A higher bit depth (or digitization) will result in enough grey values to produce a smooth change from one tone to another. The number of bits defines the number of grey levels (shown below).

  • 8bit = 256 grey levels
  • 10bit = 1,024 grey levels
  • 12bit = 4,096 grey levels
  • 14bit = 16,384 grey levels

The image above was captured with a lower bit depth. Notice the lack of smooth changes in the image, as well as the lack of some fine details.
